The movie begins with the introduction of the three main characters, Moe (Chris Diamantopoulos), Larry (David DeLuise), and Curly (Sean Hayes), who are struggling to make a name for themselves as comedians. They land a gig at a theater in New York City, but their performance is met with disaster. The trio then sets out to find their long-lost mother, Mary (Jane Lynch), who abandoned them as children.

Left on the doorstep of an orphanage run by nuns, Larry, Curly, and Moe grow up finger-poking, nyuk-nyuking, and woo-wooing their way to uncharted levels of knuckleheaded misadventure. Now, out to save their childhood home, only The Three Stooges could become embroiled in an oddball murder plot... while also stumbling into starring in a phenomenally successful TV reality show.

The film is structured into three distinct "episodes" that follow , Larry , and Curly from their childhood at a Catholic orphanage into adulthood.
